硒对大鼠胚胎神经细胞增殖和分化的影响

The Effects of Selenium on Growth and Differentiation of Cultured Neuron from Rat Embryo Midbrain

  • 摘要: 为探讨不同浓度硒对神经细胞增殖和分化的影响,将细胞培养物经不同浓度(0.01~10μmol/L)亚硒酸钠(Na2SeO3)处理,用MTT法检测细胞增殖率,并观察细胞形态变化和细胞分化程度.结果:低浓度硒(0.01~0.1μmol/L)能促进神经细胞增殖和分化;较高浓度硒(0.05~5μmol/L)则抑制神经细胞增殖和分化,并均呈剂量-效应关系,高浓度硒(10μmol/L)引起细胞毒性.提示低浓度硒有助于分化期大鼠胚胎神经细胞增殖和分化.

     

    Abstract: The effects of selenium on growth and differentiation of primary cultured neuron from thirteenth rat embryo midbrain were investigated.The cultures of cells were exposed to 0 01~10 0μmol/L Na2SeO3 and observed the changes of proliferation of neurons using MTT stain. The degree of differentiation and morphological changes of neurons were examined by means of scanning electron microscope and phase-contrast microscope.The results showed that low Se concentration (0.01~0.1μmol/L) can stimulate proliferation and differentiation of cultured neurons with a relationship of does-effect response.While the higher Se concentration (over 1.0μmol/L) can inhibit it, highest Se concentration(10μmol/L) can cause cytotoxicity.Low Se concentratation is helpful to growth and differentiation of neurons from rat embryo.
