含铅和无铅汽油尾气排出颗粒有机提取物的细胞膜毒性研究

Comparison of Membrane Toxicity between Two Extracts Using ULG and LG

  • 摘要: 为揭示无铅汽油能否减少汽车尾气颗粒有机提取物的细胞膜毒性,在发动机台架试验上分别使用市售90#含铅、无铅两种汽油,检测其尾气中颗粒物排放量并提取它们吸附的有机物,利用体外培养中国仓鼠肺细胞(CHL),通过检测细胞培养液中乳酸脱氢酶(LDH)及细胞内K+,Ca2+浓度分析细胞内外物质的交换,观察不同汽油排出尾气颗粒有机提取物的细胞膜毒性.结果显示无铅汽油的使用能减少颗粒吸附有机物对细胞膜的损伤,一定程度上降低了它们的细胞膜毒性.

     

    Abstract: The usage of unleaded gasoline(UL G)was initiated in Shanghai on Oct.1997.In order to explore whether the ULG can decrease the cell membrane toxicity.Two kinds of gasoline including ULG and leaded gasoline(LG)were tested on Engine Dynamometer under the condition of idle and medium-duty.The gasoline exhaust particulates(GEPs)were collected and the or ganic matter adsorbed was extracted.And In vitro Chinese hamster Lung(CHL) cells culture was carries out.Cells were treated with extr acts separately to detect the release of cytosolic enzyme Lactate Dehydrogenase(LDH)and the fuidity of K+,Ca2+.The results demonstrated that the concentration of LDH in medium and Ca2+ insids cells increased and that of K+ inside cells decreased accordingly.Doseresponse relatio nships were also found,which suggested GEPs had the ability of damag ing cell membrane and could cause the leakage of LDH,K+ out of cell and the inflow of Ca2+ into cell.
