The Screen on Insland LEE Containing eae Gene from Special Fragment of EHEC O157∶H7

  • 摘要: PCR扩增EHECO157:H7eae基因的特异片段并制成杂交探针,采用斑点杂交和酶切方法从自构建的基因文库中筛选不含SmaⅠ的LEE片段.结果筛选到长约23kb的LEE片段.表明筛选片段长度与预测不符,可能存在变异,需进一步证实.

     

    Abstract: To screen pathogenicity island LEE containing eae gene. Methods Special frament of EHEC O157:H7 was amplified by PCR and tagged as probe by DIG.Meanwhile,a fragment of LEE containing no SmaⅠsite was obtained through dot blothy-bridization and enzyme digestion.Result An about 23kb-long fragment of LEE was generated.Conclusion The lenghth of screened fragment is not in accordance with the predicted.
