慢性镉中毒肾小管上皮细胞凋亡形态学研究

Morphological Study on Apoptotic Cells of Renal Proximal Tubules Exposed Cadmium Chronically

  • 摘要: 目的 探讨慢性染镉对肾近端小管上皮细胞凋亡的影响.方法 用免疫组化细胞凋亡染色(Tunel法)及透射电镜,观察慢性镉中毒小鼠肾近端小管上皮细胞凋亡(apoptosis)的形态结构改变,并用图象分析法对凋亡细胞计数.结果 染镉后,小鼠肾近端小管上皮细胞可见较多的细胞凋亡.凋亡细胞呈单个或局灶性分布,主要有以下形态特点:(1)细胞胞体变小,胞质浓缩,嗜酸性反应增强.(2)细胞核核仁消失,染色质浓缩边集于核膜下,核呈环状,晚期核固缩、变小,颜色深染、致密度高.(3)核碎裂,形成凋亡小体.透射电镜观察,凋亡小体形态不规则,外被质膜,为电子致密团块,内含碎裂的核染色质及一些变性崩解的细胞器.经细胞计数,与正常对照组比较近端小管上皮细胞凋亡染色阳性反应细胞数量明显增多(P<0.01).结论 慢性染镉有促进近端小管上皮细胞凋亡的作用.

     

    Abstract: Objective To study effects of chronic cadmium intoxication on apoptosis of epithelial cells of renal proximal tubules.Methods The morphological and structural change of the apoptotic cells of renal proximal tubules of mice exposed cadmium chronically were observed by immunohistochemistry technique(Tunel method)and electron microscopy.The apoptotic cells were counted with image analysis.Results More epithelial cell of renal proximal tubules appeared apoptosis.Apoptotic cells are diffuse or crowded in the epithelium.The cell had the following characters:(1)The volumes of the cells became smaller,and the cytoplasm was concentrated and appeared acidophilic.(2)The nucleoli disappeared and concentrated chro2 matins distributed along the nuclear envelopes.The nucleus looked like a circlet.In later period the nuclei were found pyknosis and were became smaller and compacter.(3)The nuclei appeared karyorrhexis.The apoptotic bodies were seen in some nucle-i.They were covered by the plasma membrane and the shape of them were irregular.They had high electorn-dense and contained broken chromatins and organelles.Compared to the normal control group,the numbers of the apoptotic cells of the renal proximal tubules increased obviously(P<0.01).Conclusion The apoptosis of the cells of the renal proximal tubules were prompted when the mice were intoxicated chronically by cadmium.
