成纤维细胞生长因子对硫芥细胞毒的保护作用研究

Evaluation of Protective Effects of Fibroblast Growth Factor on Sulfur Mustard-induced Cytotoxicity

  • 摘要: 目的 评价基因重组碱性成纤维细胞生长因子(rbFGF)对硫芥诱导的细胞毒的保护作用.方法 以人表皮角质细胞株HaCaT细胞为实验模型,选用了rbFGF作为实验药物,分别用细胞显微镜下形态学变化、XTT法测细胞增殖活力、流式细胞术(FCM)测细胞膜损伤等方法来评价其防治效果.结果 和染毒对照组相比,显微镜观察显示rbFGF处理组漂浮细胞减少,而贴壁细胞增多,XTT法显示OD值显着增高,FCM显示M1值(PI荧光染色细胞百分率)减少.同时还发现硫芥染毒前1小时加入rbFGF进行预处理并不能提高防治效果.结论 rbFGF对于硫芥在HaCaT细胞模型的细胞毒作用具有明显的拮抗效果.

     

    Abstract: Objective To evaluate the protective effects of recombinant basic fibr oblast growth factor(rbFGF)on sulfur mustard(HD)induced cytotoxicity.Methods The HaCaT cell line derived from normal human epidermal keratinocytes was used as the research model.The protective effects of rbFGF on HD-induced cytotoxicity were evaluated by microscopic observation on morphologic change of cell culture,measurement of cell viability by XTT method and cellular membrane damage by flow cytometry(FCM).Results Compared with the HD-control group,the rbFGF treating group was found that:the number of floating cells was lesser while the number of cells sticking to the wall was more showed by microscopical examination; the OD value was significantly higher examined by XTT method;the M1 value(the percentage of PI fluor escing cells)was lesser showed by FCM.But 1 hour-pretr eatment of rbFGF failed to provide more prophylactic and ther apeutic effects.Conclusion rbFGF was found to provide significant protective effects against HD-induced cytotox icity in HaCaT cell line.
