胎儿小脑组织中Cu、Fe、Mn、Zn的测定研究

Study on Determination of Cu, Fe, Mn, Zn in Fetal Cerebellum

  • 摘要: 目的 建立胎儿小脑组织中生物必需微量元素Cu、Fe、Mn、Zn的分析方法.方法 样品经HNO3+H2O2微波消解体系消解后,用电感耦合等离子体发射光谱法ICP-AES,同时测定Cu、Fe、Mn、Zn含量.结果 元素的检出限分别在0.0007mg/L~0.03mg/L之间;线性良好,线性相关系数均≥0.999;重现性好,变异系数RSD>2%;准确度高,回收率在93.7%~116.1%之间,用猪肝GBW08551及人发GBW07601标样质控,分析结果与标准值吻合.样品分析结果显示Cu、Mn呈正态分布,Fe、Zn呈正偏态分布.结论 本法快速灵敏,精密度及准确度均符合要求,结果令人满意.

     

    Abstract: Objective To establish the method for determination of biological essential trace-elements Cu,Fe,Mn,Zn in fetal cerebellum.Methods Closed-vessel microwave digestion with HNO3+H2O2 was used for sample preparation and the digested analyze was directly analyzed for the determination of Cu,Fe,Mn,Zn by ICP-AES.Results The detection limit of each element was between 0.000 7mg/L and 0.03mg/L.Standard linear correlation r≥0.999.Standard deviations were below 2%.Recoveries of spiked natural samples were of the range 93%-117%.Two standard reference materials namely "Pig Liver GBW08551" and "Human Hair GBW07601" were used as quality control standards and the analysis results conformed well to the certified values.The frequency of results for Cu and Mn showed normal distribution,and the frequency of results for Fe and Zn showed skewness distribution.Conclusion The proposed method was proved to be simple,rapid,accurate and reliable,and ithas been applied to the analysis of the samples with satisfactory results.
