甲基叔丁基醚对大鼠小脑γ-氨基丁酸影响

Effect of methyl tertiary butyl ether on γ-amino-butyric acid in cerebral of rat

  • 摘要: 目的 研究甲基叔丁基醚(Methyl tertiary butyl ether,MTBE)对大鼠小脑皮质神经递质γ-氨基丁酸(γ-GABA)的影响,探讨MTBE对中枢神经系统的急性系作用机制.方法 54只健康成年SD大鼠,体重180~220g,随机分为9组,腹腔注射染毒,染毒剂量分别为0,700,1050,1400mg/(kg·bw).观察各染毒剂量及染毒后0.5,1,2,4,8h时MTBE对大鼠小皮质神经递质GABA的影响.结果 MTBE对小脑皮质神经递质GABA的作用其阳性产物相对灰度值结果显示,不同剂量700,1050,1400mg/(kg·bw)MTBE染毒后小脑皮质神经递质GABA相对灰度值随着染毒剂量增加而增加,存在剂量-效应关系;MTBE对小脑皮质GABA的影响在染毒后0.5h时GABA表达开始增加,1.0h达到高峰,2.0h时开始减弱,8.0h时GABA表达恢复到正常水平.结论 MTBE对中枢神经系统急性毒作用与神经递质GABA含量的改变有关.

     

    Abstract: Objective To explore the acute toxic mechanism of methyl tertiary butylether(M T BE),in the cerebellar cortex by detecting the change of the neuro transmitter C-amino-but yric acid(GABA).Methods 54 adult SD rats were randomly divided into control and eight experiment groups.The MTBE exposed doses were 700,1 050,1 400mg/(kg·bw) and the control group was used normal saline 1 400 mg/(kg·bw)respect ively injected by abdo minal cavity.Then the relative grey value of GABA expressio n was mensurated after exposed MTBE in 0.5,1.0,2.0,4.0,8.0 h in cer ebellar cortex.Results The expression of the GABA in cerebellar cortex was inceased when exposed MTBE700,1 050,1 400 mg/(kg·bw).The expression of the GABA in cerebellar cortex was changed in the different time(0.5,1.0,2.0,4.0,8.0 h)after being exposed to M T BE(1 050 mg/(kg·bw)).T he expression of the GABA began to increase in 0.5 h,reached peak in 1.0 h,began to reduce in 2.0 h,and was recovered to normal level in 8.0 h.Conclusion The acute toxic effect of MT BE in central nervous system is relevant to the neurotransmitter GABA.
