5-HTR4和GABRG2基因多态性与精神分裂症关系

Relationship between gene polymorphism of 5-HTR4 as well as GABRG2 and schizophrenia

  • 摘要: 目的 探讨5-羟色胺4受体基因(5-HTR4)和γ-氨基丁酸-γ2受体基因(GABRG2)基因多态性与精神分裂症的关系。方法 在95个中国北方汉族精神分裂症患者和他们的健康父母组成的核心家系中,以聚合酶链式反应-限制性片段长度多态性(PCR-RFLP)方法对5-HTR4基因rs888957(C/T碱基改变)和GABRG2基因rs211014(T/G碱基改变)单核苷酸多态性(SNP)进行检测。应用基于家系的连锁不平衡方法分析基因型数据。结果 (1)5-HTR4基因rs888957及GABRG2基因rs211014基因型频率的分布均符合Hardy-Weinberg平衡;(2)传递不平衡检验(TDT)结果提示,5-HTR4基因rs888957及GABRG2基因rs211014与精神分裂症无连锁及关联;(3)UNPHASED双位点单体型分析结果显示,rs888957-rs211014单体型系统与精神分裂症无关联(P>0.05);(4)5-HTR4基因的rs888957位点等位基因分布与精神分裂症的阳性症状钟情妄想相关联(P<0.01)。GABRG2基因的rs211014位点等位基因分布与精神分裂的阳性症状冲动伤人行为相关联(P<0.05)。结论 5-HTR4基因rs888957位点及GABRG2基因rs211014位多点态性可能与精神分裂症无关,但不能排除5-HTR4基因及GABRG2基因其他SNPs与精神分裂症相关联。

     

    Abstract: Objective To investigate the association between schizophrenia and the polymorphism of 5-hydroxytryp-tamine-4 receptor(52HTR4)and gamma-aminobutyric cidreceptor,gamma-2(GABRG2).Methods PCR-restriction fragment length polymorphism(RFLP)was used to examine the C/T single nucleotide polymorphism(SNP)of 5-HTR4 and T/GSNP of GABRG2 with 95 nuclear family trios of schizophrenia.Results The genotype frequency distributions of two SNPs were in the Hard-Weinberg equilibrium.The results of TDT indicated that the allelic frequency transmitted from the heterozygote parents did not deviate 50%.The results of transmission test showed that rs888957-rs211014 haplotype system was not associated with schizophrenia.There was significantly relationship between 52HTR4 allele and delusion of love that was positive symptom of schizophrenia(P<0.01).There was significantly relationship between GABRG2 allele and aggressive behavior(P<0.05).Conclusion As far as Chinese han schizophrenia in this paper is concerned,we may not be assured the association between the polymorphism of 5-HTR4 or GABRG2 and schizophrenia,but we can not be excluded the effect of polymorphism of 5-HTR4 and GABRG2 on psychotic symptoms in schizophrenia.
