人参总皂苷对缺氧小鼠脑皮质HIF表达影响

Effect of ginsenosides on HIF expression in anoxic mices cortices

  • 摘要: 目的 探讨人参总皂苷对缺氧小鼠大脑皮质缺氧诱导因子-1α(hypoxia inducible-factor-1α,HIF-1α)表达的影响。方法 Western检测缺氧小鼠大脑皮质HIF-1α蛋白表达;RT-PCR检测缺氧小鼠大脑皮质HIF-1αmRNA表达;常规苏木素-伊红(HE)染色检测脑组织损伤。结果 人参皂苷可以明显增强缺氧小鼠大脑皮质HIF-1α蛋白表达,缺氧12,24,36 h条件下与对照组比较可分别提高14.25%,29.65%及101.76%,相应降低缺氧导致的脑组织损伤。结论 人参皂苷增强缺氧小鼠大脑皮质HIF-1α表达,可能在其保护缺氧脑组织中起着重要作用。

     

    Abstract: Objective To study the effect of ginsenosides on HIF expression in anoxic mice's cortices.Methods Westren Blot and RT-PCR were used to test HIF expression,H-Estain was used to check the damage of cortices of anoxic mice.Results Ginsenosides could significantly enhance the HIF protein expression under anoxic environment and lessen the hypoxia damage on mice cortices.Compared with the control group,hypoxia for 12,24 and 36h the expression of HIF-1α protein of ginsenosides group increased 14.25%±29.65% and 101.76%,respectively.Conclusion Enhancing the expression of HIF may play an important role in the neuroprotective effect of ginsenosides on aroxic mice's coetices.
