丙烯酰胺致大鼠肺损伤及敏感标志物遴选

Measurement of alveolar surfactants content after acrylamide-induced lung injury

  • 摘要: 目的 观察丙烯酰胺致大鼠肺损伤后肺表面活性物质(PS)中磷脂(PL)的组分(磷脂酰甘油(PG)、磷脂酰肌醇(PI)、磷脂酰乙醇胺(PE))和蛋白质含量变化,寻找丙烯酰胺损伤早期敏感特征性标志物。方法 24只大鼠气管内滴注丙烯酰胺(0.1 mg/kg,0.4 mg/kg)建立肺损伤模型。分别设12 h,2,3,5 d组。大鼠的肺组织块分别用苏木素-伊红(HE)染色,光镜观察。测定支气管液中PL组分和蛋白质含量。结果 实验组大鼠肺均可见PS层丧失连续、均匀的绒状结构,脱落入肺泡腔,以12 h组较明显。PS中PL含量趋于增加,以12 h组较明显(55.61±9.97)。PS中蛋白质含量最高,以2 h组较明显(180.4±22.5)。结论 丙烯酰胺损伤后PS的质与量的变化能较特异的反映肺损伤。测定PL含量可判断肺早期损伤。PL中的磷脂酰甘油(PG)和磷脂酰肌醇(PI)可能是丙烯酰胺损伤早期敏感特征性标志物。

     

    Abstract: Objective To observe the change of composition of PL and content of protein in PS after acrylamide damage of lungs.Methods The model of rat lung injury was made by intratracheally instilling acrylamide(0.1 mg/kg,0.4 mg/kg).24 rats were divided into four groups:122hour group,22day group,32day group,52day group.PS of each group were examined by electron microscope.Content of PL,composition of PL and content of protein of each group were determined respectively.Results Structure changes of PS were found in rats'lungs of experimental groups and 122hour group was more apparent.Content of protein in PS was the highest in 122hour group(55.61±9.97)and 122hour group was more apparent(180.4± 22.5).Conclusion Morphologic change and alternation of quality and quantity of PS after anatoxin2induced pulminary injury specifically reflect the process of lung damage.Content of PL and PI is a simple and feasible marker when lung injured by acrylamide in early stage in rat.
