PFOS致大鼠肝脏氧化损伤及对脂褐质含量影响

Oxidative damage of perfluorooctane sulfonate on lipofuscin levels in rat livers

  • 摘要: 目的探讨全氟辛烷磺酸(Perfluorooctane,sulfonate,PFOS)经口亚慢性染毒致大鼠肝脏氧人损伤及对脂褐质(Lipofuscin,LF)含量的影响。方法24只雄性SD大鼠按体重随机分为4组,各染毒组饮水中PFOS浓度分别为1.7,5.0,15.0 mg/L。连续染毒90 d后,观察大鼠肝脏脏器系数;测定大鼠血清PFOS浓度、肝脏丙二醛(MDA)、LF含量和总抗氧化能力(T-AOC)。结果各染毒组与对照组大鼠体重差异无统计学意义(P>0.05);各染毒组血清PFOS浓度均显著高于对照值(P<0.05);5.0和15.0 mg/L染毒大鼠肝脏重量分别为(19.5±1.3),(20.4±4.8)g,显著高于对照值(14.2±3.4)g(P<0.05)。肝脏脏器系数分别为(4.20±0.18)%,(4.95±0.56)%,显著高于对照值(3.54±0.41)%(P<0.05)。肝脏MDA含量分别(2.38±0.92),(1.82±0.48)nmol/(mg·prot),显著高于对照值(0.87±0.11)nmol/(mg·prot)(P<0.05)。T-AOC分别为(1.33±0.20),(1.35±0.18)U/(mg·prot),显著高于对照值(1.08±0.17)U/(mg·prot)(P<0.05);15.0 mg/L染毒组大鼠肝脏LF含量为(2.89±0.15)μg/ml,显著高于对照值(2.68±0.09)μg/ml(P<0.05)。结论PFOS可致大鼠肝脏氧化损伤并增加肝脏脂褐质含量。

     

    Abstract: ObjectiveTo evaluate oxidative damage of perfluorooctane sulfonate(PFOS)subchronic exposure on lipofuscin (LF) levels in rat livers.MethodsTwenty-fourmale SD rats were randomly divided into 4 groups The rats were administrated with PFOS at concentrations of 1.7, 5.0, 15.0mg/L, in drinking water, respectively.After 90 days of exposure,liver somatic index, seiwn PFOS concentration, levels of maglonydiadehyde (MDA),LF and total antioxidant capacity(T-AOC) in liver were deteunfined.ResultsAfter 90 days exposure, body weights of rats in dosage groups showed no significant difference with those in control group(P>0.05).In 5.0, 15.0mg/L, PFOS dosage groups, liver weight and liver somatic index, serum PFOS concentration, levels of MDA and T-AOC in liver increased(P<0.05).In 15.0 mg/L PFOS dosage group, levels of LF were significantly higher than that of the control group.ConclusionIt is suggested that PFOS could induce the oxidative damage and increase the levels of LF in rat livers.
