Abstract:
ObjectiveTo investigate the β-amyloid precursor prote in(APP)gene changes induced by sodium sulfite.
MethodsRT-PCR was used to study mRNA changes of APP and Western-blotting was used to detect the prote in level of APP in human embry onic kidney cell line 293(HEK 293)treated with sodium sulfite.
ResultsRT-PCR results showed that treatment of 0.039-10 mmol/L Na
2SO
3 could not change the mRNA level of APP;Western-blotting results showed that 10 mmol/L Na
2SO
3 could decrease the protein level of APP in the cells significantly.On the other hand,0.039-2.5 mmol/L Na
2SO
3 could increase the prote in level of APP inside cells.Semi-quantity analysis showed that APP prote in levels were 1.596,1.630,1.556,and 1.446 times,respectively,when compared to that of the control.
ConclusionNa
2SO
3 can cause significant increase of APP prote in level in HEK 293 and itmaybe related to Alzheimers' disease.But the change is not due to the change of mRNA of APP.
