隐孢子虫感染小鼠补骨脂治疗效果评价

Therapeutic effect of Fructus psoraleae and dihydroartemisinin on cryptosporidiosis in mice

  • 摘要: 目的 观察补骨脂、双氢青蒿素及二者配伍治疗隐孢子虫感染小鼠前后的肠壁黏膜组织病理变化和T细胞表面的CD28分子表达水平,探讨这3组中药对隐孢子虫病防治的疗效。方法 60只昆明种小鼠随机分成正常组A、阳性对照组B、补骨脂治疗组C、双氢青蒿素治疗组D以及二者配伍治疗组E。经1和2周药物治疗后,观察各组小鼠肠壁黏膜组织病理变化和T细胞表面的CD28分子表达水平。结果 经过1和2周的治疗,药物治疗组小鼠的粪便卵囊量与阳性对照组比较,均明显减少(P<0.05);小肠病理学显示,小肠上皮处于修复之中。在经药物治疗后,所有治疗组的CD28分子表达明显高于阳性对照组B1(P<0.05),补骨脂治疗组C2最高(64.01±5.39),明显高于双氢青蒿素治疗组D2(P<0.05)。结论 补骨脂和双氢青蒿素能抑杀隐孢子虫,其中双氢青蒿素杀虫效果明显,而补骨脂主要通过调节外周血CD28分子,以及CD4+与CD3+T细胞比例,二者通过协同作用,参与宿主免疫应答和炎症反应过程,最终修复炎症反应。

     

    Abstract: Objective To investage the effects of Fructus Psoraleae and dihydroartemisinin on expression of CD28,pathological change of intestine in mice infected with Cryptosporidium parvum.Methods Cryptosporidium parvum infection was induced by hypodermic injection of dexamethasone in mice,twice a week for 4 weeks.Sixty mice were divided into 5 groups in random:A(normal group),B(control group),C(therapeutic group of Fructus psoraleae),D(therapeutic group of dihydroartemisinin),E(therapeutic group of the two agents combination).According to the period of therapy(1 or 2 weeks),therapeutic groups were subdivided into A1,A2,B1,B2,C1,C2,D1,D2,and E1,E2.Results Compared with the control group,the number of Crypatosporidium parvum oocysts in feces were decreased obviously in all treatment groups(P<0.05).The pathological damage of intestinal epitheliums were renovated after therapeutic treatment.Proportions of CD28 T lymphocytes in peripheral blood in all treatment groups were higher than those of infected control groups(P<0.05).Conclusion Fructus psoraleae and dihydroartemisinin are effective in the treatment of cryptosporidiosis in mice.Dihydroartemisinin can remarkably eliminate Cryptosporidium parvum.Fructus psoraleae can up-regulate peripheral blood CD28 and proportion of CD4+ and CD3+ T cells.The two agents participate in the host immune response and inflammatory reaction with synergic effect and ultimately restore inflammatory responce.
