家蝇幼虫分泌型抗菌肽分子鉴定

Molecular identification of antibacterial piptide from Musca domestica larvae secretion

  • 摘要: 目的 十二烷基磺酸钠对家蝇幼虫分泌型抗菌肽进行分子鉴定。方法 对纯化所得的家蝇幼虫分泌型抗菌肽使用SDS-聚丙烯酰胺凝胶电泳(SDS-PAGE)及肽指纹图谱方法进行分子鉴定。结果 SDS-PAGE结果表明,纯化得到的2个抗菌组分,主要成分的分子量在14.157 kDa左右,另一组分的分子量为7.579 kDa,但蛋白含量明显低于上一组分;分别将这2个抗菌肽分子命名为家蝇幼虫分泌型抗菌肽Ⅰ和抗菌肽Ⅱ,抗菌肽Ⅰ经肽指纹图谱分析,精确分子量为14.264 kDa,其与溶菌酶序列有一定同源性,序列覆盖率为20%,但两者生物学特性不同,推测为不同蛋白质。结论 从家蝇幼虫分泌物中分离得到的抗菌肽与溶菌酶有一定同源性,但生物学特性有差异。

     

    Abstract: Objective To identify the antibacterial peptide from the larvae of Mu scadomestica with tris-tricine sodium dodecyl sulfate polyacrylam idegel electrophoresis(SDS-PAGE)and pepitde finger printing method.Methods The antibac terial peptide was separated and purified form the secretion of Mu scadomesticala rvae.The molecular weight of the peptide was determined with tric-tricine SDS-PAGE;the an tibacterial piptide molecule was identified with peptide finger printing method.Results Two active compounds were isolated and named as secreting type an tibacterial peptideⅡ(AP-Ⅱ) and secreting type antibacterial peptideⅡ(AP-Ⅱ).The irmolecular weihgt are 14.157 kDa and 7.579 kDa by SDS-PAGE,respectively,and the accurate molecular weight of AP-1 is 14.264 kDa determined with mass spectrum technique.There is a homology between AP-1 and lysozyme of the aminoacid sequence.Fraction of sequence coverage is 20%.But they are different pioteins according to their different biochemical charac teristics.Conclusion The AP-1isolated from secretion of Musca donestica larvae has some homology and different biochemical charateristics withly sozyme.
