铅对仔鼠学习记忆及其海马组织TNF-α表达影响

Effects of maternal lead exposure on learning and memory ability and expression of TNF-α in hippocampus of offspring mice

  • 摘要: 目的 探讨母体铅暴露对仔鼠海马CA 1区肿瘤坏死因子-α(TNF-α)表达影响.方法 小鼠自孕第1 d开始经饮水染铅(0.3、1.0、3.0 g/L)至仔鼠出生后21 d断乳,对照组饮用不含铅的蒸馏水.每组随机取仔鼠10只于出生后第7、14、21 d检测血铅和海马铅;出生后第21 d,用Y型迷宫测定学习记忆能力,用免疫组化法测定海马CA 1区TNF-α蛋白表达.结果 铅暴露组生后7、14和21 d的仔鼠血铅、海马铅水平均明显高于对照组(P<0.05).Y型迷宫显示,各染铅组出生后21 d的仔鼠,T 1、T 2和T 3值均低于同期对照组,差异均有统计学意义(P<0.05).免疫组化显示,与对照组(0.079±0.012)比较,低、中、高剂量染铅组仔鼠海马CA 1区TNF-α免疫组化阳性反应物面密度(0.154±0.013、0.197±0.018、0.201±0.015)明显增加,差异均有统计学意义(P<0.05);中、高剂量染铅组仔鼠海马CA 1区TNF-α阳性反应物平均灰度值(102.36±36.76、86.36±31.52)明显低于对照组(223.41±52.21),差异有统计学意义(P<0.05).结论 母体孕哺期铅暴露,引起仔鼠海马CA 1区TNF-α高表达,学习记忆能力下降.

     

    Abstract: Objective To explore the effects of lead exposure in pregnant mice on the expression of tumor necrosis factor-α(TNF-α) protein in hippocampus of mouse pups.Methods Lead acetate was administered to pregnant mice in drinking water at the dosages of 0.3,1.0,and 3.0 g/L.Each group composed of 10 mice.At the 7th,14th,and 21st day after the birth,the lead contents in blood and hippocampus of the offspring were determined.At the 21st day,the learning and memory ability of the offspring mice were tested by Y-type maze,and the expression of TNF-α protein in hippocampus of the offspring was determined by immunohistochemistry assay.Results At the 7th,14th,and 21st day,the lead levels in blood and hippocampus of the offspring mice of the lead exposure groups were higher than those in control group(P <0.05).The results of Y-type maze demonstrated that T1,T2,and T3 for 21-day old offspring in the three lead exposed groups were significantly decreased compared with those of the control group(P < 0.05).The results of immunohistochemistry assay showed that the area densities of TNF-α positive immune reaction in CA1 area of mouse pups in 0.3,1.0,and 3.0 g/L lead acetate exposure groups(0.154±0.013,0.197±0.018,0.201±0.015) were higher than that of the control group(0.079±0.012,P < 0.05 for all),and the averages gray values of TNF-α in CA1 area of mouse pups in 1.0 and 3.0 g/L lead acetate exposure groups(102.36±36.76,86.36±31.52) were lower than that of the control group(223.41±52.21,P <0.05 for all).Conclusion Maternal lead exposure may induce accumulation of lead in the offspring,enhance the expression of TNF-α protein in hippocampus of pups and result in learning and memory damages.
