PM2.5空气污染物对大鼠肺部炎症细胞因子表达影响

Effect of ambient air PM2.5 on expression of inflammatory cytokines in lung tissue and bronchoalveolar lavage fluid in rats

  • 摘要: 目的 分析PM2.5空气污染物致大鼠呼吸系统炎症损伤过程中肺部炎症细胞因子表达变化。方法 90只Wistar大鼠随机分成12组,每组6~8只。染尘染毒组大鼠经气管注入PM2.5混悬液染尘,第2天动式染毒,对照组经气管注入生理盐水并吸入正常空气;ELISA法检测大鼠肺泡灌洗液(BALF)中炎症细胞因子含量,实时荧光定量PCR技术检测大鼠肺组织中炎症细胞因子mRNA表达。结果 与对照组比较,染尘染毒1、7、30 d 染尘染毒组大鼠BALF中细胞因子白细胞介素-4(IL-4)、IL-6、肿瘤坏死因子-α(TNF-α)含量及大鼠肺组织中IL-4、IL-6、TNF-α mRNA表达量均表现为先升高后下降趋势,而γ干扰素(IFN-γ)变化不明显,但呈持续下降趋势。结论 在PM2.5空气污染物致大鼠呼吸系统炎症损伤过程中,体液免疫应答占主导地位,Th1/Th2平衡失调,PM2.5空气污染物不能刺激Th1保护性免疫反应;巨噬细胞清除PM2.5空气污染物动力不足。

     

    Abstract: Objective To explore the effect of particulate matter ≤2.5 microns in aerodynamic diameter (PM2.5) on expressions of inflammatory cytokines in respiratory damage in rats.Methods Ninety Wistar rats were randomly divided into 9 experimental groups exposed to low-,moderate-,and high-dose of PM2.5 (via trachea injection of PM2.5 saline suspension) and polluted air (one day after PM2.5 exposure) containing low-,moderate-,and high-content of sulfur dioxide (SO2),nitrogen dioxide (NO2),and carbon monoxide (CO) and 3 control groups treated with normal saline and normal air;the treatments were conducted 4 hours a day continuously for 1,7,and 30 days.The inflammatory cytokines in bronchoalveolar lavage fluid (BALF) were detected with enzyme-linked immunosobent assay (ELISA) and the mRNA of inflammatory cytokines in lung tissues were detected with real-time fluorescent quantitative PCR.Results Compared with those in the control groups,the expressions of interleukin-4 (IL-4),interleukin-6 (IL-6),and tumor necrosis factor-α (TNF-α) in BALF and their mRNA in lung tissues were increased first and then decreased in experimental groups during the treatments of PM2.5 and air pollutants;while the level of interferon gamma (IFN-γ) showed no significant change,with a declining trend.Conclusion Humoral immunity is a major response in the process of respiratory system injury caused by airborne PM2.5 in rats with unbalanced T helper 1 (Th1)/T helper 2(Th2)ratio.PM2.5 could not activate protective immune response of Th1 and macrophages in lung could not eliminate PM2.5 effectively.
