Cytotoxicity effect of arsenic on HaCat cells and protective effect of N-acetylcysteine

      目的   研究亚砷酸钠(sodium arsenite, NaAsO2)对人皮肤角质形成细胞系(HaCaT)的毒性作用及N乙酰半胱氨酸(Nacetylcysteine, NAC)对砷毒性的拮抗作用。
      方法   NaAsO2单独作用于HaCaT细胞24h或用NAC预处理后, 加入NaAsO2继续作用24h, 用AlamarBlue还原法检测细胞活力。
      结果   < 1μmol/L的NaAsO2单独作用于细胞24h后, AlamarBlue还原率增高(P < 0.05), 10 μmol/L以上的NaAsO2则使AlamarBlue还原率显著下降(P < 0.01);用NAC预处理24h后再加入NaAsO2, 20 μmol/L组AlamarBlue还原率与NaAsO2单独作用相比显著增加(P < 0.05)。
      结论   低浓度NaAsO2刺激细胞增殖, 高浓度具有细胞毒性; NAC可减轻砷的细胞毒性, 细胞内的还原型谷胱甘肽(GSH)对砷的细胞毒性起保护作用。

     

    Abstract:
      Objective   To study the cytotoxicity of sodium arsenite(NaAsO2)on keratinocytes(HaCaT)and the protective effect of N-acetylcysteine(NAC).
      Methods   The HaCaT cells were treated with sodium arsenite alone or pre-treated with NAC for 24 hours, followed by the addition of sodium arsenite.AlarmarBlue assay was used to detect the proliferation of the cells.
      Results   The reduction rate of AlarmarBlue increased when the HaCaT cells were treated with 0.001 μmol/L-1 μmol/L of NaAsO2 alone(P < 0.05).When pre-treated with NAC for 24 hours, the reduction rate increased significantly in the 20 μmol/L group compared with the group treated with 20μmol/L of NaAsO2 alone(P < 0.05).
      Conclusion   Low levels of NaAsO2 stimulate the proliferation of HaCaT cells while high levels inhibit it.NAC can alleviate the cytotoxicity of asenic, which indicate that the cellular GSH exert the important protective effects on the cytotoxicity of arsenic.
