预热对白血病患者胸腔渗出细胞H2O2的影响

Effects of pro-heat on sensitivity of K562 cell to hydrogen peroxide

      目的   探讨预热对肿瘤细胞过氧化氢敏感性的影响及其机制。
      方法   将白血病患者胸腔渗出细胞(K562)在40℃预热不同时间, 诱导细胞热应激蛋白70(HSP70)高表达, 用RT-PCR检测细胞HSP70mRNA水平; 将细胞暴露于0.1, 0.2, 0.4, 0.8mmol的H2O216h, 用噻唑蓝(MTT)比色法检测细胞活力, 观察预热对不同水平H2O2敏感性的影响; 保持H2O2浓度为0.2mmol不变, 观察不同预热时间对细胞H2O2敏感性的影响。
      结果   40℃预热后细胞的HSP70含量升高, 在预热120min后达到最高, 以后缓慢下降, 可维持10h以上; 预热细胞对不同浓度H2O2的敏感性均显著低于未预热细胞; 对于相同浓度的H2O2而言, 预热120min的细胞对敏感性最低。
      结论   预热可以引起细胞HSP70表达增高, 降低细胞对H2O2的敏感性。

     

    Abstract:
      Objective   To study the effects of pre-heat on the sensitivity changes of cells to the TNF-α and its mechanism.
      Methods   The K562 cells were pre-heated in 40℃ for different time, then were exposed to the hydrogen peroxide, with different concentrations of 0.1, 0.2, 0.4, 0.8 mmol/L, for 16 hours; at the same time the cell vitalities of different time were detected by MTT with constant exposure to 0.02 mmol/Lhydrogen peroxide.
      Results   The HSP70 could be induced by preheat, and it went its top after 120 min pre-heat, and the sensitivity of non-heat cell to hydrogen peroxide was lower than that of pre-heated cells.
      Conclusion   Pre-heat could induce the HSP70 expression and inhibit the sensitivity of cell to hydrogen peroxide.The mechanism maybe involved with the over expression of HSP70.
