艾滋病病毒1型流行株分子流行病学调查

Molecular epidemiological investigation on HIV-1 epidemic strain

      目的   了解广东省HIV1亚型的流行规律及其与国际参考株的同源性。
      方法   应用套式聚合酶链反应(PCR)对108例广东省HIV1感染者淋巴细胞富集液的核酸样品进行扩增, 并使用ABI377型测序仪对扩增产物测序后, 对其外膜蛋白(ENV)基因C2V3段的核酸序列进行比较分析。
      结果   108份血样中, 44份为AE重组亚型, 与国际参考株01AE.TH.90.CM240最近, 基因离散率为(8.644±1.273)%, 组内离散率为(7.302±4.639)%; 40份为07BC重组亚型, 与国际参考株CN.97.C54A最近, 基因离散率为(2.163±1.313)%, 组内离散率为(3.388±1.686)%; 14份为08BC重组亚型, 与国际参考株97CNGX9F最近, 基因离散率为(2.757±2.027)%, 组内离散率为(3.728±1.971)%; 10份为B亚型, 其中9份为泰国B′, 与国际参考株RL42最近, 基因离散率为(5.687±2.232)%, 组内离散率为(5.879±2.542)%; 1份为欧美B, 与国际参考株US.86.JRFL最近, 基因距离为8.50%。
      结论   广东省HIV1流行株以AE和07BC重组亚型为主, 也存在08BC重组亚型、泰国B亚型和欧美B亚型。

     

    Abstract:
      Objective   To investigate the subtype of HIV-1 prevelent in Guangdong province.
      Methods   HIV-1 proviral DNA from buffy coat of 108 infectors in Guangdong was amplified by nested PCR.The C2-V3 regions of HIV-1 ENV gene were sequenced directly from the PCR product and were analyzed.
      Results   The 108 infectors were confirmed to be infected with five HIV-1 subtype or Circulating Recombinant Form(CRFs): AE(n=44), 072BC(n=40), 082BC(n=14), B'(n=9)and B(n=1).The AE group showed the highest homology to 01AE.TH.90.CM240 strain isolated in Thailand, and the genetic distances was(8.644±1.273)%.The 07-BC group showed the highest homology to CN.97.C54A strain, and the genetic distances was(2.163±1.313)%.The 08-BC group showed the highest homology to 97CNGX-9F strain, the genetic distances was(2.757±2.027)%.The B′group showed the highest homology to r142 strain isolated in Thailand, and the genetic distances was(5.87±2.232)%.THe B subtype showed highest homology to US.86.JRFL strain isolated in USA, and the genetic distance was 8.50%.The innergroups distance among AE, 07-BC, 08-BC, B'group were(7.302±4.639)%, (31388±11686)%, (3.728±1.971)%, (3.728±1.971)%, respectively.
      Conclusion   AE and 07-BC were main epidemic subtypes in Guangdong province.08-BC, B' and B were also circulating in Guangdong province.
