Orthogonal experiment of single cell gel electrophoresis assay using minimal peripheral blood samples

  • 摘要:
      目的   研究外周微量血单细胞凝脉电泳技术的最佳实验条件。
      方法   采用四因素三水平的正交表L9(34)对电泳电压、电泳时间、解旋时间、铺胶层数四因素作正交试验。
      结果   解旋时间为最主要影响因素, 其次分别为铺胶层数、电泳时间和电泳电压; 并且各因素下的第一水平为最佳水平。
      结论   外周微量血单细胞凝胶电泳技术的最佳实验条件为: 解旋20min、铺胶3层、电泳20min、电泳电压20V。

     

    Abstract:
      Objective   To optimize the experimental conditions for single cell gel electrophoresis assay(SCGE).
      Methods   Orthogonal experiment(4 factors×3 levels)were carried out: unwinding time, gel layer number, electrophor esis time, and electrophoresis voltage.
      Results   Unwinding time was the main factor, followed by the gel layer number, electrophoresis time, and electro phoresis voltage.Furthermore, number one level under four factors was optimum level.
      Conclusion   Optimized experimental conditions consisted of 20-minute unwinding time, gel of 3 layers, 20-minute electrophoresis time, and 20 volt electrophoresis voltages.
