感染血吸虫小鼠肝脏中一氧化氮合酶动态探讨

Co-expression of iNOS and HSP-70 in livers of mice infected with schistosoma japonicum

      目的   探讨感染血吸虫小鼠肝脏中一氧化氮合酶(iNOS)与热休克蛋白(HSP)-70表达的动态变化。
      方法   日本血吸虫尾蚴经皮肤感染小鼠后第0, 21, 28, 38和45d分别取其肝脏, 用RTPCR半定量法检测iNOS表达的动态变化; 用Westernblot方法检测HSP70表达的动态变化。
      结果   感染后第21d起能检测到iNOS的表达, 感染后第38d达峰值, 第45d时表达水平有所下降; HSP70在未感染小鼠肝脏内有微弱的表达, 在感染后21d的肝脏其表达显著增强, 38d达峰值, 45d后表达下降。
      结论   血吸虫感染可诱导iNOS和HSP70在肝脏的协同表达, 且两者表达的动态变化趋势一致。

     

    Abstract:
      Objective   To study the time-course change in expressions of iNOS and HSP-70 in the livers of mice infected with schistosoma japonicum.
      Methods   The livers of mice infected with schistosoma japonicum were collected before infection and at day 21, 28, 38, 45 post infection.Total RNA of livers were extracted, time-course change of the mRNA expression of iNOS and protein expression of HSP-70 were detected by RT-PCR and Western-blot, respectively.
      Results   The mRNA expression of iNOS was detectable in the livers of all infected mice from day 21 post infection other than uninfected mice.The expression was increased at day 28 and reached peak at day 38, while decreased slightly at day 45.Protein expression of HSP70 were detectable in the livers of both infect ed and uninfected mice, the mode of time-course change was as same as that of iNOS.
      Conclusion   Co-expression of iNOS and HSP-70 in the liver could be induced by the infection of schistosoma japonicum, both iNOS and HSP-70 had the same mode of time-course change in expression.
