摘要:目的 探讨SOD活性及SOD基因表达与衰老的相关性。方法 选用快速老化模型鼠(SAM-P/8)进行试验, 采用生物化学及RT-PCR方法, 从亚细胞及分子水平上测定衰老过程中丙二醛(MDA), 超氧化物歧化酶(SOD)及其基因表达的变化情况。结果 SAM-P/8肝组织中SOD活性随着年龄的增加而逐渐降低; 而MDA含量随年龄的增加逐渐增高, 有显著性差异(P < 0.05)。不同月龄SAM-P/8肝细胞中Cu-Zn SOD基因表达随年龄的增加Cu-Zn SOD mRNA逐渐降低(P < 0.01)。结论 SOD活性与SOD基因表达的增龄变化有明显的相关性, SOD基因转录活性的降低可能是导致SOD活性降低的重要原因, 表明SOD在衰老过程中扮演着重要角色。Abstract:Objective To explore the changes on activity and expression of SOD in the progress of senescence.Methods Senescence-accelerated mouse p/8(SAM-P/8)was used in the study.Biochemical assay and Reverse Transcription Polymerase Chain Reaction(R T-PCR)were adopted to measure changes occurred on SOD activity and gene expression in the progress of senescence.Results Activity of SOD decreased significantly with aging(P < 0.05), while the level of MDA increased significantly with aging(P < 0.05).And the level of Cu-Zn SOD mRNA derived from the liver of SAM-P/8 decreased with aging(P < 0.01).Conclusion Activity of SOD gradually decreased with aging.The decreasing expression of Cu-Zn SOD gene was an important reason for SOD's decreasing activity.It could be inferred that SOD played an important role in the progress of aging.
