流感病毒裂解过程中的形态变化规律

  • 摘要:
      目的   研究流感病毒在裂解剂作用下不同时间的形态变化及对病毒亚单位结构形态上的影响, 掌握流感病毒在裂解剂作用下的裂解规律。
      方法   将纯化后的流感病毒抗原按0.5%~0.6%比例加入裂解剂Triton N101, 分别在0.5, 1, 2, 3, 3.5, 4, 5, 6h取样, 用有膜电镜载网粘取样品, 用2%磷钨酸(PTA)负染, 电镜下观察。
      结果   流感病毒裂解可大致分为基质蛋白(Mprotein, MP)包膜膨胀阶段, 神经胺酸酶(Neuraminidase NA)、血凝素(Hemagglutinin HA)、脂质双层(Lipidbilayer LB)脱落阶段, 基质蛋白包膜破裂核衣壳(Nucleocapsid NP)溢出阶段、基质蛋白溶解阶段。
      结论   采用震荡措施可使病毒间裂解进程趋于同步。

     

    Abstract:
      Objective   To realize the transformation shape of influenza virus under the action of split, and the action about the virus substructure shape. To learn split way.
      Methods   To draw split influenza viruse specimen of A1, A3 and B subtypes at different time, and observe under electron microscope. The split solvent is Triton N-101.
      Results   The splitting of influenza virus can be divided into four stages. The stage includ M protein (Mp) membrane swell neuraminidase (NA) hemagglutinin (HA) lipid bilayer (LB) break away phase, Mprotein (Mp) membrane disrupt, nucleocapsid (NP) spill and Mprotein dissolved phase.
      Conclusion   The method of vibration makes split better.
