鸡冠花黄酮类对成骨细胞增殖和TGFβ1的作用

Effects of soybean ethanol etract on osteoblastic cell proliferation and TGFβ1

  • 摘要:
      目的   探讨鸡冠花黄酮类化合物对体外培养大鼠颅骨成骨细胞增殖、分化和转化生长因子-β(TGFβ1)表达的影响作用, 为临床骨质疏松症的预防和治疗提供参考。
      方法   提取新生大鼠颅骨成骨细胞制作体外培养模型, 然后用噻唑盐(MTT)法测定鸡冠花黄酮类化合物对体外培养成骨细胞的增殖作用, 氨基安替吡啉测酚法测成骨细胞内碱性磷酸酶(ALP)活性, SP免疫组化法研究鸡冠花黄酮类化合物对成骨细胞TGFβ1表达的影响。
      结果   鸡冠花黄酮类化合物可以促进体外培养大鼠成骨细胞的增殖, 并显著性促进成骨细胞分化。对第3代成骨细胞TGFβ1表达进行图像分析, 培养3d, 鸡冠花黄酮类化合物剂量在0.5以上组TGFβ1表达阳性, 培养5d, 所有实验TGFβ1均表达阳性促使成骨细跑合成TGFβ1的免疫组化阳性表达。
      结论   鸡冠花黄酮类化合物可以提高成骨细胞的增殖和分化, 并有促进分泌TGFβ1的功能, 对于预防骨质疏松症的发生有重要的参考价值。

     

    Abstract:
      Objective   To explore the effects of flavonoid of celosia cristata on proliferation, differentiation and excretion of TGFβ1 in vitro cultured osteoblasts.
      Methods   Osteoblastic cells from newborn rat calvarial was cultured.Osteoblastic cells proliferation was measured by MTT, activity of ALP was observed.SP immunohistochemistry was used to measure the effects of flavonoid of celosia cristata on expression of TGFβ1 in vitro cultured osteoblasts.
      Results   The flavonoid of celosia cristata had the effects of stimulating the proliferation, differentiation of osteoblasts and increasing the expression of TGFβ1 in vitro cultured osteoblasts in rat.
      Conclusion   The flavonoid of celosia cristata can stimulate the proliferation, differentiation and the expression of TGFβ1 in calvarial osteoblastic cells.
