小鼠肺泡巨噬细胞的提取、纯化及活性检测

Extracting, purifying and activity testing of pulmonary alveolar macrophages of mice

  • 摘要:
      目的   探讨从小鼠提取肺泡巨噬细胞(PAM)的可行性。
      方法   采用支气管肺泡灌洗法从小鼠、大鼠肺内采集游离细胞, 用贴壁法提取肺泡巨噬细胞, 并以酸性磷酸酶(ACP)法和吞噬鸡红细胞实验检测巨噬细胞的纯度。
      结果   单位肺组织2种鼠的肺泡巨噬细胞收获量无显著性差异。
      结论   采用灌洗法提取小鼠肺泡巨噬细胞是一种简便易行的办法。

     

    Abstract:
      Objective   To investigate the feasibility of extracting pulmonary alveolar acrophages(PAM)from mice.
      Methods   Free pulmonary alveolar cells from mice and rats were collected by bronchoalveolar lavage fluid(BALF), and PAM were extracted by pasting.The purity of PAM were measured by acid phosphatase(ACP)assay and chicken red blood cell (CRBC)phagocytisis assay.
      Results   There were no significant difference of PAM in quantity between two groups.
      Conclusion   It was a simple and convenient assay of extracting PAM from mice by BALF.
