Vc/FeSO4诱发红细胞膜脂质过氧化模型的建立

The Foundation of Erythrocyte Membranes Lipid Peroxidation Model Induced by Vc/FeSO4

  • 摘要: 通过体外实验,对Vc/FeSO4诱发的红细胞膜脂质过氧化(LP)发生条件及反应历程进行了探讨。结果表明:在37°C水浴45分钟内,红细胞膜蛋白浓度为0.75mg/ml,5.0μmol/LFeSO4,pH值7.4的5mmol/L磷酸盐缓冲液的条件下,较低浓度Vc(小于1.0mmol/L)时可促进红细胞膜脂质过氧化过程,但高浓度Vc(大于1.0mmol/L)则抑制红细胞膜脂质过氧化过程。Vc浓度为0.25mmol/L时,红细胞膜脂质过氧化产物丙二醛(MDA)于反应后45分钟内迅速上升,90分钟接近峰值,以后趋于平衡。揭示:一定比例的Vc/FeSO4可有效地激发红细胞膜LP.本研究初步建立了红细胞膜LP模型。

     

    Abstract: The condition and thetime course of lipid peroxidation(LP) induced by Vc /FeSO4 in human erytrocytem embranes invitrow as studied. The results revealed that, when human erythoccytem embranes(0.75mg/ml protein) were incubated with 5μmol/L PBS buffer for 5 minutes, Vc, at low concentration(<1.0 mmol/L) stimulate lP, but at high concenration(>1.mmol/L), inhibated LP. The content of malonaldehyde(MDA) rised rapidly after additon of 0.25mmol/L Vc and 5.0μmol/L FeSO4, reached maxinal level in 90minutes, very little additional LP occured over the next two hours. The result suggested that in 45 minutes, Vc /FeSO4 the appropriate scale could induced effectively erythrocytem embranes LP. A model induced by Vc /FeSO4 was founded preliminarily in our experimnet.
