Transcutaneous electrical resistance test as an alternative method of skin corrosion test in rat

  • Objective To establisha method of transcutaneous electrical resistance(TER)test and to evaluate the feasibility of the method in rat.Methods TER value and histopa thological changes of normal rat skin discs were observed to evaluate the feasibility of the isolated skin fixing aPPARγatus assembled.Tenchemicals were tested according to the Organization of Economi Cooperation and Development Guide line for Testing of Chemicals No.430.Statistical analysis was conducted to determine the relevance of the TER test compared with the data of in vivotest.Results The barrier function of normal skin discs fixed in the aPPARγatus were stable with in 24 hours and the TER value variation curve swere repea table.The morphological structure of the skin discs was intact with in 24 hours.The TER test was concordant with in vivotest for the ten reference chemicals(Mcnemar,P=0.500;Kappa=0.615,P=0.035)with the sensitivity of 100% and specificity of 66.7%.Conclusion The isolated skin fixing aPPARγatus could be used for TER test.The TER test could a lternate skin corrosion test for testing corro sive chemicals.
