Joint effect of specific circular RNA and overweight on diabetes: a hospital-based case-control study

  •   Objective  To investigate the expression of circular RNAs (circRNAs) in peripheral blood lymphocytes and joint effect of circRNAs expression and overweight on diabetes for providing references for the prevention and control of diabetes.
      Methods  We conducted questionnaire interviews, physical examination and laboratory detection among 233 diabetic cases and 937 controls recruited at two general hospitals affiliated to Fujian Medical University in Fujian province during April 2017 and October 2018. Real-time quantitative PCR was used to detect the expression of circRNAs (hsa_circ_0008507, hsa_circ_0001946 and hsa_circ_0000284) in 36 cases and 164 controls. Multivariate logistic regression model and crossover analysis were utilized to analyze the interaction between circRNAs and overweight and their effect on diabetes.
      Results  After adjusting for confounding factors such as gender and age, overweight (odds ratio OR = 1.402, 95% confidence interval 95% CI: 1.020 – 1.929), high expressions of hsa_circ_0008507 (OR = 2.661, 95% CI: 1.099 – 6.447) and hsa_circ_0001946 (OR = 3.790, 95% CI: 1.140 – 12.602) were risk factors for diabetes incidence. Further analyses revealed that the participants with both overweight and high expression of hsa_circ_0008507 (OR = 6.122, 95% CI: 1.485 – 25.237) and the participants with both overweight and high expression of hsa_circ_0001946 (OR = 4.523, 95% CI: 1.148 – 17.816) had increased risk of diabetes. There were no multiplicative and additive interaction between overweight and high expressions of hsa_circ_0008507 and hsa_circ_0001946 based on interaction analysis (P > 0.05).
      Conclusion  The expression of hsa_circ_0008507 and hsa_circ_0001946 may be associated with diabetes incidence; the high expression of hsa_circ_0008507 and hsa_circ_0001946 combined with overweight could increase the risk of diabetes.
