Study on Non-agglutination McAb to Human Red Blood Cell Antigen and its Application

  • Objective To obtain non-agglutination McAbs to human red blood cell antigens for constructing antibody reagents with two-function,which could be used in clinical diagnosis in lab.Methods BALB/c mice were immunized with human O group erythrocutes.Lymphocyte hybridoma technique was used to make McAb.Results Hybridoma cell lines(7A1,7A2,1H7,7H3)which could stably secrete McAbs against human red blood cell antigens were obtained.The isotype assay demonstrated that all the light chains of the McAbs were the K type and all the heavy chains were IgG1.The McAbs could specifically recognize the human red blood cell antigens on A,B,AB,O groups red blood cells from 200 donators respectively by Cooma's test and no cross-reaction was found with red blood cells from ten kinds of animals respectively.The application in full blood hemagglutination test for detecting hemorrhagic fever with renal syndrome(HFRS) was studied further.Conclusion The non-agglutination McAb(7Al) to human red blood cell antigen could be used to construct the antibody reagent with two -function.
