Effects of grape Procyanidins on proliferation of hepatocytes in vitro and vivo

  •   Objective   To study the effectof GPC on the prolifer ation of normal hepatocytes and the cells damag ed byethanol.
Methods   ① Normal primary mouse hepatocytes and the cells damag ed byethanol were cultrued with grape procyanidins(GPC)of different concentrations.Methyl thiazolyl tetrazolium(MTT)assay was used to analyze the cell proliferation.②GPC was given tonormal mouse and the ones with liverinjuried byethanol acutively through mouth.Cell proliferations of 3 h and 16 h were analyzed by the same way as above.
Results   In vitro the proliferations of normal control and the ethanol group were 0.438±0.040 and 0.365±0.024 respectively. Those of the 50 mg/LGPC protective group and control group were 0.541±0.026 and 0.603±0.060 respectively. In vivo, the proliferation rates of the 150 mg/LGPC protective group were 0.095 higher than thatof the ethanol one.Thatof the 150 mg/LGPC control group was 0.030 higher than thatof the normal control one.All differences abo ve were sig nificand(P < 0.05).
Conclusion   GPC can inhibitthe damage of ethanol on proliferation of hepatocytes, and promote proliferation of normal cells.
