Effect of SDS on resonance light-scattering spectrum of histone and DNA complex

  •   Objective   To study the effect exerted by sodium dodecyl sulfate(SDS)on the resonance lig h-tscatter ing based on histone and DNA complex and to explore the orderliness of inter action in the three compounds.
      Methods   Histone, SDS and DNA were mix ed into the Tris-HCl buffer solution(pH 7.5).Common spectrofluoromenter was used to measure the resonance light-scattering intensity of the mix ture with same excitation and emission waveleng th.
      Results   Scan spectrum show ed that the max imal light-scattering wavelength was 302nm, and the optimum measurement for ligh-tscattering intensity was at pH 71 5.The ligh-tscattering sig nal was strong in the range of 0.02~0.15 mmol/LSDS.
      Conclusion   Lo w dose of SDS improv ed the resonance ligh-tscattering intensity of histone and DNA.Otherwise, high dose of SDS urged the segregation of histone and DNA, socut down thereso nance light-scatter ing intensity greatly.
