Effect of arsenic on expression of NF-κB in HaCaT cells

Abstract

Abstract

Objective To evaluate effects of sodium arsenite on expression of nuclear transcription factor-JB(NF-JB), reactive oxygen species(ROS)and catalase(CAT)in HaCaTcells.
Methods ROS was detected by flow cytometry(FCM), activity of CAT was detected by ultraviolet velocity method and western blot technique was used to detect expression of NF-JB protein.
Results The level of ROS increased when NaAsO₂ was above 2.5 μmol/L in a dose-dependent fashion.The activity of CAT and the expression of NF-JB decr eased significantly from 5 μmol/L grour(P < 0.05).
Conclusion Arsenic induces oxidative stress in HaCaTcells and inbibits ex pression of NF-½B protein.
