Inhibitory effect of ebselen and GSH on peroxidation in rats

  •   Objective   To inv estigate inhibitory effect of Ebselen and glutathione(GSH)on perox idation of Alcohol-dependent rats.
      Methods   Serum glutathione peroxidase(GPx)activity and lipid peroxide(LPO)content and malo ndialdehyde (MDA)of alcohol-dependent rats were detected in end of 3 weeks afterexposure, and ratio of GPx activity to MDA was applied to index the inhibitory effect of peroxidation.The ratio of each ebselen-exposure group with different concentration substracted the ratio of positive control group, the result was divided by the ratio of ebselen-exposure group and a percentage was gotten which represented the efficiency of inhibition.
      Results   Ebselen and GSH were administerd simultaneously and a synergistic effect could be invesgated when the concentration of Ebselen and GSH varied from 0.5mg/kg to 10mg/kg and 5~10mg/kg, respectively.The efficiency of inhibition increased from 40.0% to 65.7% (Par < 0.01), which displayed a dose-response relationship(r=0.693, P < 0.01).When ebselen of different concentration(5~10 mg/kg)was administered singly, the inhibitory effect could also be obtainde which varied from 36.7% to 45.3%.
      Conclusion   Ebselen can display antiox idative effect with the co-factor GSH, and other pathways may also exertin the mechanism of antiox idant effect of ebselen.
