Analysis on gene expression profile of laminin receptor

  •   Objective   To investigate the gene expression changes related to laminin receptor, and to give the basis data for the study of carcinog enesis of anti-BPDE.
Methods   The transient transfected 16HBE cell line with 67LR1 gene was constructed by using lipofectin protocols. The expression products of transfected genes was analyzed by semi-quantity RT-PCR.The gene expression patterns in the transient transfected 67LR1 cell line and the non-transfected parental cell line were detected by cDNA microarray which representing 8 046 different genes.
Results   The transient 16HBE cell lines with 67LR1 was successfuly constructed.Analysis of the gene expression profile indicated that there were 295 genes expressed differently between transfected 67LR1 gene lines and the non-transfented parental cell line.Among these changed genes, 145 genes were up-regulated, and 150 genes down regulated. These significantly changed genes involved in cell signal transduction genes, tumo-rrelated genes, immune-related genes and protein synthesis related genes.
Conclusion   The analysis of 67LRl-related gene profile alterations indicate that 67LR1 may be induces not by an alteration in the expression of a single gene and a single signal pathway, but by the accumulation of alteration in the expression of several genes and complex signal transduction network.
