Effect of Schistosoma japonicum sj23 gene transfer on function of dendritic cell in vitro

Abstract
Objective To explore the effect of 23kDa membrane protein of Schistosoma japonicum(Sj23)gene transfer on the function of dendritic cell(DC)in vitro.Methods Sj23 gene was introduced into DC by liposome-mediated gene transfer met hod.DCs were transfected with recombinant plasmid pcDNA3-Sj23.The ability of DC to take an antigen was determined by flow cytometry(FCM).The mixed lymphocy tereaction(MLR)was used to detect the effect of Sj23 gene-transferred DC on stimulating allogenetic Tlymphocyte.Results Sj23 was expressed at high level in the Sj23 gene-transferred DC.Compared with pcDNA3 transfer red DC and untreated DC,the ability of antigen intake of Sj23 gene-transfer red DC decreased and Sj23 gene-transferred DC had strong potential to stimulate proliferation of allogenetic Tlymphocy te.Conclusion Sj23 gene-transfer can promote DC activation and enhance the biologic activity of DC.
