Effect of Schistosoma japonicum sj23 gene transfer on function of dendritic cell in vitro

  • Objective To explore the effect of 23kDa membrane protein of Schistosoma japonicum(Sj23)gene transfer on the function of dendritic cell(DC)in vitro.Methods Sj23 gene was introduced into DC by liposome-mediated gene transfer met hod.DCs were transfected with recombinant plasmid pcDNA3-Sj23.The ability of DC to take an antigen was determined by flow cytometry(FCM).The mixed lymphocy tereaction(MLR)was used to detect the effect of Sj23 gene-transferred DC on stimulating allogenetic Tlymphocyte.Results Sj23 was expressed at high level in the Sj23 gene-transferred DC.Compared with pcDNA3 transfer red DC and untreated DC,the ability of antigen intake of Sj23 gene-transfer red DC decreased and Sj23 gene-transferred DC had strong potential to stimulate proliferation of allogenetic Tlymphocy te.Conclusion Sj23 gene-transfer can promote DC activation and enhance the biologic activity of DC.
