Detection of fluorescent material in arsenic-resistant cells with LSCM

  • Objective To detect the fluore scentmaterial with in arsenic-resistant cells by lase rscanning confocalmicro scope(LSCM).Methods The arsenic-resistant cells were incubated with fluorescent dyes Rhodamine-123 for 30 minutes.The cells of test group were coincubated with Verapamil and NaSO2,respectively.The cells of control group were merely administrated with Rhodamine 123.Fluo rescence miage acquisition of Rhodamine-123s' dynamic series were executed with LSCM,and the concentration fluorescence intracell at different time(12,24,36,48,64h)was recorded.Results After fluo rescence staining for 48h,the fluorescence intensity of the test group was 38.94±0.3648 and 38.153±0.533 for Verapamil and NaSO2 treamtent.The intensities were significantly higher than that of the synchrono us control group.Conclusion The LSCM could be used to detect fluorescent material in arsenic resistant cells tmiely and quantitively.
