Study on signal pathways of cyanidin-3-glucoside in association with inhibiting inflammation and carcinogenesis by gene microarray

  • Objective To investigate the effect of cyanidin-3-glucoside (C3G)on gene expression relevant to signal pathway in THP-1 macrophage by DNA microarray for screening signal pathways of cyanidin-3-glucoside in associstion with inhibiting inflammation and carcingensis.Methods THP-1 macrophages were treated with 100 μmol/L C3G for 12 hr,then the expression levels of RNA transcipts were assessed using human signal transduction pathway finder microarray analysis,and some of the results were confirmed by semi-quantitative RT-PCR.Results The results showed that 15 genes in 7 signal transduction pathways were differentially regulated in the THP-1 macrophages at 12 hr post C3G-treatment.Five genes including TP53,NFKBIA,RBP2 were upregulated and 10 genes including FOS,NOS2A,PTGS2 were downregulated.These genes encode a host of proteins as the important targets in signal transducition pathways involved in inflammatiry response and carcinogensis.Conclusion Anthocyanins might inhibit the inflammatory response and carcinogensis via divergent signal pathways such as mitogenic pathway,Wnt pathway,p53 pathway,NFκB pathway,and JAK/STAT pathway.
