Effect of repeated fasting/refeeding on NUCB 2 expression in hypothalamus of rats

  • Objective To explore the expression of leptin and nucleobindin-2(NUCB2)after repeated fasting/refeeding in rats with high fat diet.Methods The rats were treated with repeating fasting/refeeding for 6 weeks.Then the model group was switched to high fat dietevery day(high fat model group),and the control group was designated as high-fat and common-fat control groups.At the end of 12 weeks,the Lee's index and body-fat content of the rats in diffenent groups were compared.The concentrations of serum lipid and leptin were measured.The expression of NUCB2 in hypothalam us was detected with mimunoh isto chemical method.Results The serumleptin level of model group(3.18±0.54 ng/m L) was sign ificantly higher than that of control group(2.44±0.45 ng/mL,P<0.05).The expression of NUCB2 was also significantly higher than that of control group(P<0.01).The Lee's index,body fat content, serum total cholesterol,trigly ceride,low density lipoprotein,and leptin level of high fat model group were significantly lower than those of high fat control group whereas significantly higher than those of common fat control group(P<0.05).While the expression of NUCB2 was significantly lower than that of high fat and common fat control groups(P<0.01).Conclusion The repeated fasting/refeeding feeding pattern can decrease the obesity induced by high fat diet, and also reduce the leptin resisitance,but cause obvious decrease in NUCB2 expression in hypothalamus of obese rats.
