Antagonistic effect of sodium ferulate on oxidized low-density lipoprotein

  • Objective To study the antagonistic effect of sodium ferulate(SF) on oxidized low-density lipoprotein(Ox-LDL) in human umbilical vein endothelial cells.Methods Human umbilical vein endothelial cells(HUVEC) were cultured in vitro.Gene chip technology was used to analyze the change of interleukin(IL)-1β gene expression in HUVEC after the treatments of Ox-LDL,SF+Ox-LDL,respectively.The IL-1β mRNA was measured with real time PCR(RT-PCR) and IL-1β protein by enzyme-linked immunosorbent assay(ELISA).Results The expression of IL-1β gene was up-regulated by 2.4 folds after the induction of Ox-LDL in HUVECs compared with that of the control.While the expression of IL-1β gene was down-regulated by 2.6 folds after the induction of SF+Ox-LDL in HUVEC compared with that of in the group of Ox-LDL.The results of RT-PCR and ELISA showed that both the expression of IL-1β mRNA and IL-1β protein increased significantly in Ox-LDL group compared with that of the control.The expression of IL-1β protein decreased obviously while the expression of IL-1βmRNA didn't change significantly in SF+Ox-LDL group compared with that of the control.Conclusion Ox-LDL could induce the expression of IL-1β,which could be inhibited by sodium ferulate.Meanwhile,sodium ferulate could also down-regulate the expression of IL-1β so as to reduce the damage of IL-1β in endothelial cells.
