Effect of curcumin on expression of adiponectin and resistin mRNA in 3T3-L1 adipocytes

  • Objective To explore the effect of curcumin on the expressions of adiponectin and resistin mRNA in 3T3 L1 adipocytes,and to analyze possible mechanism of curcumin in the prevention of diabetes at cellcular level.Methods Curcumin at concentrations of 2.5,5,10,15,and 20 μmol/L was added in 3T3-L1 preadipocytes during induced-differentia tion for six days and 5,10,and 20μmol/L in mature 3T3-L1 adipocytes for 24 hr,respectively.Then the expressions of adi ponectin and resistin mRNA in 3T3-L1 preadipocytes during induced-differentiation and mature 3T3-L1 adipocytes were de tected with RT-PCR.Results The expression of adiponectin mRNA increased and resistin mRNA decreased in 3T3-L1 preadipocytes treated with curcum in compared to 3T3-L1 preadipocyte swithout curcum in during induced-differentiation.The expression of adiponectin mRNA increased and resistin mRNA decreased in mature 3T3-L1 adipocytes treated with curcum in compared to that of in mature 3T3-L1 adipocytes without curcumin.Conclusion The increase of adiponectin expression mRNA and the decrease of resistin mRN A in 3T3-L1 adipocytes may be one of mechanisms of curcumin in the prevention of diabetes.
