Effect of HCV-F gene on transcription activation of c-myc promoter

Abstract
ObjectiveTo observe the effect of hepatitis C virus F(HCV-F)gene on transcription activition of c-myc promoter and to explore the role of HCV-F on patho genesis of hepatocarcino genesis.MethodsThe luciferase reporter plasmids of c-myc promoter were constructed and cotransfected in to HepG 2 cells with HCV-F and HCV-C expression plasmids.The transactivition of c-myc promoter was a ssayed by luminometer.ResultsHCV-C exhibited a distinct promotion activity to c-myc promoter,with a seven folds increase of c-myc promoter transactivity compared to that of the control group.HCV-F showed less promotion activity and enhanced luciferase activity about three folds increse of c-myc promoter.At a range of 2 to 10 g,the enhancement of c-myc promoter was observed to be dose dependent only with HCV-C not HCV-F.ConclusionHCV-Fexhibits less promotion activity for c-myc promoter than HCV-C,which does not posses all functions of HCV-C protein.
