Effects of various selenocompounds on production of SEPP and GPx in HepG2 cells

  • Objective To examine stimulative effects of various selenocompounds on the production of selenoprotein P(SEPP) and glutathione peroxidase(GPx)in HepG2 cells.Methods The cultured HepG2 cells were divided into a control group, low- (0.01μmol/L), midium- (0.1 μmol/L), and high-dose (1.00 μmol/L)groups treated with different selenocompounds, including sodium selenite(Na2SeO3), selenomethionine(SeMet), and methyl selenocysteine(MeSeCys).Fourty-eight or 72 hours after the treatments, the cell culture supernatants and homogenates were collected for the detections of SEPP and GPX with double-antibody sandwich enzyme-linked immuno sorbent assay(ELISA).Results Compared with the control group, the concentrations of SEPP(115.78±1.70, 140.73±0.97, and 114.92±1.46 pg/mL)increased significantly in high-dose Na2SeO3, SeMet, and MeSeCys groups 48 hours after the treatment(all P<0.01)and those of GPx(490.52±5.40, 643.26±5.40, and 603.35±6.40 U/L) also increased significantly(all P<0.01).Compared with the other two selenocompounds(Na2SeO3 and MeSeCys), SeMet showed the highest stimulative effect(P<0.05).No significant difference in the stimulative effect was observed between the treatments of 48 and 72 hours.ConclusionAmong the three selenocompounds, SeMet has the highest stimulative effect on productions of SEPP and GPx in HepG2 cells.
