Research on spectinomycin and ciprofloxacin resistance and resistant gene in neisseria gonorrhoea

  •   Objective   To discuss the resistance and mechanism of spectinomycin and ciprofloxacin.
      Methods   The neisseria gonorr hoea strains were isolated by Cholocate Poly Vitex Agar in sexually transmitted diseases(STDs)clinic, and identified by Biomerieux gonoco ccusidentify system from France.Minimal inhibitory concentration(MIC)of spectionomycin and ciprofloxacin was detected by agar diluted method.PCR method was used to detect nucleotide transferring enzymeant gene and special gene segment in 16S ribosomal RNA(16S rRNA)and gyrA among resistant and mild sensitive strains, and to analyze DNA sequence.
      Results   In all 95 Neisseria gonorrhoea strains, 94.7%(90 straims)was spectinomycin sensitive; 4.2% (4 strains)was mild sensitive and 1.1%(1 strain)was resistant; 100% was ciprofloxacin resistant.Nucleotide transferringen zymeant gene was negative.In the spectinomycin resistance strain, a gene mutation(CT)in the No 1192 position was found.20 strains of Neisseria gonorrhoeae had been associated with mutations in the gyrA protein of the DNA gyrase.There were four types of alter ation in the gyrA.
      Conclusion   Spectinomycin is still the first choice for gonor rhoea care, but some resistant strains appear.The resistance is related to the mutation of 16SrRNA.Ciprofloxacin isn't applicable to curegonorrhea.
