Effect of DEHP on DNA damage of mice embryo fibroblast (3T6) using single cell gel electrophoresis

  •   Objective   To determinate the damage of DNA, the degree of DNA damage and the dose-effect of mice embryo fibroblast(3T6)caused by DEHP using single cell gel electrophor esis(SCGE).
      Methods   Using the mice embryo fibroblast not treated with DEHP as the negative control group, 80μmol/L H 2O 2 treated mice embryo fibroblast as the positive control group, the different concentration DEHP exposed to mice embryo fibroblast was as four dose groups(62.5, 125.0, 250.0, 500.0 μg/ml), DNA damage was detected by SCGE.
      Results   The different degree of DNA damage between groups treated with DEHP and the negative group was significant, and when the concentration of DEHP increased, the damage of the DNAin the mice embryo fibroblast was aggravated, having dose-effect.
      Conclusion   DEHP can cause DNA damage to mice embryo fibroblast and the degree of DNA damage was aggravated with the increase of DEHP.
