Influence of vitamin C on oxidative stress induced by cigarette smoke solution in rat lymphoeytes

  •   Objective   To learn how vitamin C(VC)influences oxidative stress of rat lymphocytes induced by cigarette smoke solution(CSS).
      Methods   8×10-3 cig/ml CSS was added to rat lymphocytes pretreated with or without VC, then intracellular reactive oxygen species(ROS)was detected by using 2', 7'-dichlorofluorescein diacetate(DCFH-DA), DNA damage was evaluated by comet assay, and lipid peroxide(LPO)and superoxide dismutase(SOD)activity were measured as well.
      Results   In contrast to CSS-treated cells pre-incubated with 50μmol/L VC, ROS, L PO level and DNA damage increased, and SOD activity decreased significantly in lymphocytes treated with CSS only.
      Conclusion   In rat lymphocytes, oxidative stress caused by CSS could be prevented by 50μmol/L VC effectively.
