Objective To explore the changes on activity and expression of SOD in the progress of senescence.
Methods Senescence-accelerated mouse p/8(SAM-P/8)was used in the study.Biochemical assay and Reverse Transcription Polymerase Chain Reaction(R T-PCR)were adopted to measure changes occurred on SOD activity and gene expression in the progress of senescence.
Results Activity of SOD decreased significantly with aging(P < 0.05), while the level of MDA increased significantly with aging(P < 0.05).And the level of Cu-Zn SOD mRNA derived from the liver of SAM-P/8 decreased with aging(P < 0.01).
Conclusion Activity of SOD gradually decreased with aging. The decreasing expression of Cu-Zn SOD gene was an important reason for SOD's decreasing activity. It could be inferred that SOD played an important role in the progress of aging.
