Effect of pleurotes sapidus on expression of p53 protein expression of tumor cell

  Objective   The anti-tumor mechanism of pleurotes sapidus on 4 kinds of tumor cell lines(Q₃, MGC-803, Hela, SPC-A-1)was studied.

  Methods   Four tumor cell lines were cultured in vitro.The expression of p53 was investigated by FACS techniques after treated by water extract or ethanol extract from pleurotes sapidus with doses 10^-1-10^-3g/ml.

  Results   Experimental study of MTT and HT were used to determine the antitumor effects of pleurotas sapidus on inducing apoptosis of cancer cells.The incidence rates of apoptosis were remarkably increased after treated 12 hours and 24 hours by ethanol extract(0.1 g/ml)and water extract(0.1 g/ml).The effects of ethanol extract and water extract on expression of p53 gene related to apoptosis were examined by FACS technique.The expression of p53 protein in the groups of ethanol extract and water exract after treated 12 hours and 24 hours were significantly increased.

  Conclusion   The anti-tumor effect of pleurotas sapidus was evident, and its mechanisms was probably involved in the regulation of apoptosis by inducing the gene expressions relating to apoptosis, and up-adjustment improving apoptosis gene expression(p53).
