Study on polymorphisms of TNF in patients with HBV infection in Shenyang

  •   Objective   To evaluate the effects of polymorphisms of TNF-A-238, -308 in peripheral blood moonuclear cells(PBMCs)on susceptibility of HBV and prog nosis of patients infected with HBV in Shenyang area.
Methods   Using PCR, dots blotand DNA sequencing to study the polymorphisms of TNF-A-238, -308 in PBMCs of patients with HBV infections(n=106)and controls(n=23).
Results   There was no significantdifference between do ts blotand DNA sequencing(P > 0.05).No-238A/A positive case was detected in all the samples using dots blot, 3.9% of-238A/G cases were found, 3.9% of-308A/G and 33.3% of-308G/G in all samples. There was no significantdifference of distribution of-238A/G in each group(P > 0.05).Butbetween liver cirrhosis, chronic hepatitis and carriers ithad statistical significant(P < 0.05).To the distribution of-308A/A and A/G, there was no significantdifference between each group(P > 0.05).
Conclusion   The results of do ts blotand DNA sequencing were co herent. The plymorphisms of TNF-A-238, -308 were notrelated with HBV in- fection and polymorphisms of TNF-A-308 was notassociated with the extentof diseases, however the incidence of liver cirr ho- sis increased in patients with TNF-A-238G/G if the person was infected with HBV.
