Detect Human Papillomavirus in Cervical Cancer by UsingPolymeras Chain Reaction Ligated with Digoxigenin Labeled Probe Hydrization 

  • To explore and eff ect ive method to detect human papillomavirus(HPV)in cervial cancer.Polymerase chain reaction(PCR) ligated with digoxigenin labled probe hydridization was used to detect human papillomavirus in cervical cancer.Digoxigenin labeledprobe could be used to determine the polymerase chain reaction product of human papillomavirus.The positive rate of humanpapillomavirus in 36 case of cervical cancer was 66.7%, 69.4%,69.4% respectively by using one set of consensus primer throughagarose gel elect rophoresis, PCR ligated with Southern blot hybridization and PCR ligated with dot blot hybridization.The positive rate of human papillomavirus was 72.2%(26/ 36), 75.0%(27/36), 63.2%(12/ 19),(P< 0.05), respectively by usingthree different sets of consensus primer, the total positive rate was 91.7%(33/36).The total positive rate of human papillomavirus in 27 cases of cervical cancer with β-Globin gene was 90.0%(24.27).The method to detect human papillomavi rus by using polymerase chain reaction ligated with digoxigenin labled probed could be applied to the molecular epidemiology study of cervical cancer.The positive rate might be influenced by using different sets of consensus primer.
